动物物种的繁衍和遗传信息的传递依赖于生殖细胞。在哺乳动物中，雄性生殖干细胞－精原干细胞具有干细胞的基本特征：自我更新与分化，是研究成体干细胞的经典模型，我们的研究围绕雄性生殖细胞和早期胚胎细胞在发育过程中如何在时间和空间上通过调控细胞中的蛋白合成以及蛋白组的维持调节和改变细胞的生物学功能这一问题，揭示细胞在动物发育生长过程中命运变化的分子机制，并为解析相关人类疾病的发生机制提供实验和理论基础。组织干细胞生长状态的平衡是维持组织功能、延缓器官衰老和促进组织损伤修复的基础，因此相关分子机制的研究在干细胞和再生生物学中具有广泛的意义。主要研究内容包括：
1. 精原干细胞自我更新与分化的分子调控机制：精原干细胞稳定而持续的自我更新和分化是动物持续产生精子的基础。在前期研究中，我们利用转基因小鼠模型分离、纯化了小鼠精原干细胞，并对其基因表达特征做了比较性分析。相比于精原干细胞前体细胞和分化后的生精细胞，精原干细胞具有其特异的基因表达特征和调节信号因子。解析它们在精原干细胞自我更新与分化中的作用机制将帮助我们深入理解精原干细胞如何维持生殖细胞的发育和功能、生殖系统功能的稳定和相关生殖疾病的发生机制。通过小鼠遗传学、生物化学和细胞生物学等实验，我们希望解析精原干细胞中蛋白合成调节因子参与调控精原干细胞生长的作用机制，包括相关RNA结合蛋白、信使RNA代谢和蛋白翻译调控因子的作用以及它们对精原干细胞自我更新、分化和衰老的影响。
2.精子细胞变形期中精子特异蛋白的表达调控：小鼠精子细胞发育可分为三个阶段：有丝分裂、减数分裂和细胞形态变化。初级精母细胞在完成减数分裂后进入变形期，通过一系列细胞形态的变化，包括：细胞核浓缩、顶体和尾部鞭毛结构的形成等，最终产生具有生物学功能的精子。由于细胞核中染色质的浓缩，单倍体精子前体细胞具备有限的基因转录能力，细胞变形期所需蛋白的表达多依赖于对早期存储的信使RNA的转录后修饰和蛋白翻译调控。通过对精子细胞特异的蛋白激酶A锚定蛋白AKAP3的表达和功能的研究，我们发现PKA信号通路和RNA结合蛋白在其中的潜在作用。结合生物化学及分子、细胞生物学，我们希望阐明PKA信号通路和RNA结合蛋白如何在精子前体细胞形态发生时调节信使RNA代谢、蛋白合成以及精子的功能。
3.小鼠早期胚胎发育中的细胞命运决定：在低等动物，如果蝇和线虫中，受精后胚胎发育期的细胞谱系分化（包括生殖细胞）在第一次细胞分裂时已开始，并受到受精卵中存储的母源因子的影响。这种机制是否在哺乳动物中存在，哺乳动物早期胚胎发育中细胞的命运是如何决定的？通过对小鼠着床前期胚胎细胞在分子和细胞水平上的比较、分析，我们希望解析哺乳动物早期胚胎细胞间存在的分子和功能上的差异，以及它们在动物早期胚胎发育时细胞命运决定中的作用。
