高分子材料是现代三大支柱材料之一。高分子链从分子量及分布、共聚成分及组成分布,到高分子链的拓扑结构,都直接影响着聚合物材料的性质。拓扑结构指高分子链的不同形式,如线形、星形、接枝/超支化、环形及聚合物刷等,对链拓扑结构精密合成研究可获得对高分子材料性质更深刻的认识,是本学科的基本科学问题。申请人研究高分子的组成和链结构的合成方法学,并研究这些高分子的多级结构、自组装和功能材料,进一步发展生物医用高分子材料。主要学术贡献和创新成果为:

1、星形/支化拓扑高分子的合成方法和性质

    星形聚合物具有低的溶液和本体粘度、大量端基,其结构的多样化和简单的合成方法对其性质和应用非常重要。提出多种星形聚合物的合成方法,将非烯类单体聚合物通过可控聚合转变成星形聚内酯和聚醚(Macromolecules 2004, 引用 81 次)。基于单体引发剂和大量苯乙烯共聚合,实现了“一步”简便合成星状聚合物(Macromolecules 2004, 引用 76 次),具有普适性和灵活性,解决星形聚合物合成繁琐的问题。

2、大尺寸单分子蠕虫形聚合物的简便合成

    聚合物刷和树状化聚合物单分子的长度可达数百纳米,可呈现蠕虫形貌。其形貌和尺寸与生命体系中的DNA、某些病毒具有一定的相似性,很适合研究颗粒形状在生物体系中的行为。基于可控聚合和高效成键反应,实现了长度在几十至几百纳米范围蠕虫形单分子的尺寸、结构和官能团分布的精细控制,为研究一维软纳米粒子的性质提供物质基础 (Macromolecules2005, 引用34次; Small 2006; J. Polym. Sci. Part A: Polym. Chem. 2007; 2007; 2010; Macromolecules 2007; 2009; J. Polym. Sci. Part A: Polym. Chem. 2006, 引用 63 次; Polymer 2008, 引用49次; Polymer 2012, Macromolecules 2013, 2016)。

3、聚合物纳米颗粒几何形状控制的普适方法

     提出了在反应性嵌段聚合物胶束纳米环境中进行溶胶-凝胶反应制备有机/无机杂化纳米颗粒的思想。研究了烷氧基硅甲基丙烯酸酯的可控自由基聚合(Macromolecules 2004, 引用77 次)。在此基础上合成各种两亲嵌段共聚物,在其组装体中进行凝胶交联,制备了稳定的有机/无机杂化囊泡及多种复杂结构颗粒(J. Am. Chem. Soc. 2003, 引用171次; Macromolecules 2004, 引用109 次; Angew. Chem. Int. Ed. 2004, 引用125次; Macromolecule 2007, 引用 24次)。非球形的纳米粒子具有独特的性质,但很难用简单通用的方法合成不同形状纳米颗粒。将反应性嵌段共聚物在本体微相分离再进行溶胶凝胶反应,溶剂分散后得到各种形状的聚合物纳米颗粒,微相结构决定了颗粒形状(Macromolecule 2007, 引用 43 次; 2008, 引用 32 次; Macromolecules 2012,引用86次;《高分子学报》, 2011)。

4、生物医用聚合物材料

在聚合物链结构的精密控制基础上,发展智能新型聚合物材料。通过环糊精和聚合物主客体作用制备了一种杂化碳纳米管的超分子水凝胶,具有温度响应、可注射性,为碳纳米管在生物医药中的应用提供了新原理(Macromolecules 2007, 引用 58 次);采用超分子聚轮烷作为大分子引发剂,合成出具有多级自组装行为的超分子嵌段共聚物 (Macromolecules 2008, 引用55 次);将超分子聚合物可注射水凝胶应用于纳米抗癌药物的释放,提出水凝胶-纳米药物粒子-自由药的分步释放机制(Biomacromolecules 2010,引用 45 次)。提出一种通过动态共价键进行自修复机理,在温和条件下原位对聚合物材料进行自愈合(Macromolecules 2010, 引用 244 次;年度点击前 10 位,两年中在该刊被引用前10 位)。首次实现了有机凝胶和水凝胶的宏观高强度愈合(ACS Macro Lett 2015)。发表系列相关论文(ACS Macro Lett 2012, 引用135次;Macromolecules 2012, 引用 46 次;ACS Macro Lett 2017)。

5、生物制剂纳米颗粒的功能及聚合物在自身免疫性疾病的应用

开展国际合作组建了广东省“纳米医学工程”创新团队,在载体材料设计与合成、蛋白/多肽药物纳米制剂加工、纳米制剂口服输送、纳米颗粒疫苗等研究取得了重要进展。建立了制备蛋白类药物纳米制剂的快速纳米复合技术,解决生物质药物高效和高重现加工关键问题,形成二十件余发明专利,产生技术转化前景(Biomaterials 2017; Biomaterials 2018; Nano Letters 2018, Biomaterials 2020)。提出游离核酸是类风湿关节炎发展的重要影响因素,在国际上首次提出采用阳离子聚合物通过清除游离核酸,抑制免疫反应,在动物模型上效果显著 (Nature Communication 2018; Angewandte Chemie International Edition 2019; Advanced Functional Materials 2020; Science Advances 2020)。

研究成果:
1、拓扑高分子的合成方法:研究聚合物一级链结构和拓扑结构的合成方法学,建立了具有一定普适性的简便合成星形聚合物方法,发展了制备大分子单体和类树状聚合物的新方法,提出可逐级降解、可进行拓扑链转变的星形聚合物。
2、蠕虫形聚合物的精密合成:建立了树状化聚合物和聚合物分子刷的多种合成方法学,可简便、高效、较大量合成蠕虫形纳米单分子,实现其尺寸(从几十至几百纳米)、结构和组成的控制。
3、嵌段聚合物控制纳米颗粒结构和几何形状:通过嵌段共聚物的溶液和本体自组装及选择区域交联,研究原位制备有机/无机杂化纳米粒子的方法。制备了球形、囊泡、复合囊泡等多种结构固定的纳米粒子,首次观察到内部高度折叠双分子膜的嵌段共聚物复杂囊泡,建立了可较大量制备球形、柱形、片形等形状聚合物纳米颗粒的简便普适方法。
4、新型聚合物功能材料:从分子链结构的可控发展聚合物新材料。由环糊精和聚合物的超分子作用制备了响应性、可注射、杂化碳纳米管的各种超分子水凝胶,开发其在药物输送方面的应用;提出一种简单多功能聚合物纳米药物载体体系;提出一种通过动态共价键在温和条件下对软材料进行原位自愈合的新机理。 发表SCI论文130余篇,在Macromol. Rapid Comm.和Chem. Comm.分别发表Feature Article各1篇,在Macromolecules发表Perspective Article 1篇,在《高分子学报》发表专论1篇。
